782 Characterization of mechano-biological pathways involved in bioprinted skin remodeling

Reorganization of fibrin is the first stage in matrix maturation process bioprinted skin and takes place over a few hours to days. Fibrin popular component many bioinks provisional extracellular present during wound healing. Cells embedded gels attach exert traction forces realign and/or degrade fibers, sense respond surrounding substrate mechanical feedback loop. Such cellular actions may remodel printed construct such way, rendering it no longer feasible for implantation. The goal this study understand based constructs better engineer 3D skin. Results show that fibroblasts hydrogels change viscoelastic properties networks at micron-scale, suggesting cells network mechanically interact. Rheological analysis revealed strain-stiffening pattern due non-linearly elastic forces, exhibiting when subjected large deformations. mRNA shows express elastin collagen within hydrogels.
